2. Each well of a 96-well plate can hold 300μl of liquid. We usually control the liquid volume of each well to 150μL. If there is too much liquid, it is easy to overflow and cause pollution.
3. In the experimental design, all 96 wells cannot be inoculated. Because there is less liquid in each well, it evaporates quickly in the incubator. The inoculum should be designed in the middle of the plate. Add culture medium to the surrounding wells to maintain humidity in the plate.
4. When the cells split into plates, they should be fully dispersed, and then blow out after adding a few more blanks, otherwise the cell concentration will be different
5. The wells around the 96-well plate are usually blank and do not support cells. The simple reason is the influence of temperature gradient and water evaporation. The so-called "edge effect"!
6. After adding the cells, shake the culture plate to evenly distribute the cells on the culture plate, otherwise there will be more cells around and less in the middle!
