How do you check if a pipette is calibrated correctly?

Calibration is an important aspect of pipetting accuracy, and it is essential to ensure that pipettes are calibrated correctly before use. Here are the steps to check if a pipette is calibrated correctly:

Prepare a gravimetric measurement setup: A gravimetric measurement setup involves weighing the liquid dispensed by a pipette on a balance. To set up the gravimetric measurement, weigh an empty container, then fill it with distilled water and weigh it again. The difference in weight will give you the weight of the water dispensed.

Dispense the liquid: Using the pipette you want to calibrate, dispense the liquid into the container on the balance. Make sure to follow the manufacturer's instructions for pipetting, including setting the correct volume and technique.

Weigh the liquid: After dispensing, weigh the container with the liquid in it. The difference in weight between the container with the liquid and the container with the distilled water will give you the weight of the liquid dispensed.

Calculate the error: Compare the weight of the liquid dispensed by the pipette to the expected weight. If the weight of the liquid dispensed is within the acceptable range for the pipette, then it is calibrated correctly. If the weight is outside the acceptable range, then the pipette needs to be calibrated or adjusted.

Repeat: It is important to repeat this process at least three times to get an accurate measurement and ensure consistency.
